DNA extraction from single embryosSelect either living or fixed embryos (five to ten) in a tube. Add a few chelex beads in about 50 to 100 µl water (10µl/embryo). Make sure that living embryos have been lysed, because they do not automatically lyse when boiled. Boil for a 10 minutes. Spin 5 min. Use supernatant as template for PCR analysis (1 to 5 µl in 20ul reaction volume). Material: Chelex100, Sigma C-7901, suspend 50% (v/v) in water, store at RT |
